ABO & RhD Blood Grouping Test Card (Solid – Phase Method ...
Solid-phase blood grouping (SPRCA) is a highly sensitive, automated-friendly technology that fixes antigens or antibodies to the bottom of microwells to detect ABO/Rh grouping and antibody screening. It provides 100% accuracy compared to conventional methods, appearing as a diffuse "carpet" of cells (positive) or a distinct "button" (negative). 
Key Aspects of Solid-Phase Testing 
  • Principle: In this method, reagent antibodies are immobilized on the surface of microwell plates. When patient blood is added, red cells bind to these antibodies if the corresponding antigen is present (Solid Phase Red Cell Adherence).
  • Interpretation: A positive reaction shows a "carpet" or layer of red cells spread across the bottom of the well. A negative reaction shows a concentrated red dot or "button" at the bottom of the well.
  • Advantages:
    • High Sensitivity: More sensitive than test tube methods for identifying weak antigens and antibodies.
    • Automation: Suitable for high-throughput, automated platforms (e.g., TANGO, Neo).
    • Speed & Efficiency: Allows for quick, reliable testing in high-volume laboratory settings.
  • Applications: Used for ABO/Rh forward and reverse grouping, antibody screening, and crossmatching. 
Solid-phase and gel techniques are both considered modern alternatives to hemagglutination that offer superior sensitivity and clearer results for automated systems.
